RAD (restriction site associated DNA) analysis has been used to compare the segregating nuclear DNA of plants with cytoplasm from accessions of Lolium rigidum with and without resistance to sporidesmin, a mycotoxin produced by the soil fungus Cephalosporium necator. Three accessions of L. rigidum were identified as resistant, one as moderately resistant, and two as susceptible. Outer mitochondrial (OM) DNA was amplified using primers specific to the Omt1 and Omt2 genes of OM DNA and primers to the nuclear ribosomal DNA that are distributed throughout the genome. Omt1 primers amplified DNA products from all accessions, but Omt2 only amplified DNA from resistant accessions. Outer mitochondrial DNA was distributed among resistant accessions in a manner consistent with a maternal cytoplasmic origin, thus accounting for the resistance.The Hawaii State Commission on the Status of Women wants a refund from the U.S. Education Department of more than $2.2 million it received in 2001 for its gender equality programs.
